Abstract

The anti-aging activity and mechanism of action of stachyose in vivo were investigated using the Cryptobacterium hidradenum nematode model(hereafter referred to as nematode).Methods:The antioxidant effect of stachyose was studied by measuring the body length,head swing frequency,lipofuscin,oxidative stress and thermal stress of wild-type N2 nematodes.The molecular mechanism of stachyose anti-aging was discussed by measuring the nuclear translocation of decay accelerating factor-16(DAF-16)protein of transgenic TJ356 nematode and the expression of superoxide dismutase-3(SOD-3)in CF1553 nematode.Results:It was found that stachyose not only significantly improved the head swing frequency of nematodes(P<0.001)but also significantly reduced lipofuscin deposition(P<0.001)and significantly prolonged the heat tolerance of nematodes(P<0.01).In addition,stachyose can promote the translocation of DAF-16 nucleus of TJ356 nematode and significantly increase the expression level of SOD-3 protein in CF1553 nematode.Conclusion:The antioxidant effect of stachyose was significant,and this effect could be attributed to insulin/insulin-like growth factor 1 signal pathway(IIS)signaling,which promoted DAF-16 transcription and thus stimulated SOD-3 expression,resulting in cell protection.
