摘要

Abstract

Objective:To explore the mechanism of platycodon decoction(PD)promoting Th1 cell recruitment in lung and antagonizing tumor lung metastasis.Methods:Twenty-four C57BL/6 female mice were randomly divided into a normal control group(CTRL group)and a PD medication group(PD group),and the mice in PD group received PD 2.925 g/(kg·d)by gavage.Eighteen C57BL/6 female mice were randomly divided into a normal control group(CTRL group),a B16 melanoma lung metastasis model group(B16 group),and a PD treatment group(B16+ PD group).B16 melanoma cells were injected into the tail vein of the mice in the B16 group and B16+ PD group,with PD treatment group receiving gastric lavage starting from the first 3 days.Flow cytometry was used to detect Th1 cells,its characteristic chemokine receptor CXCR3,and the expression of functional factor IFN-γ.The expression levels of CXCL9,CCL5,IFN-γ,GZMB of lung tissues were detected through RT-qPCR and single-cell sequencing.Results:Compared with the mice not treated with medication,the proportion and number of Th cells of the lung tissue were significantly increased in the mice after 10 days of treatment with PD(P<0.001);the proportion of positive expression of CXCR3 and IFN-γ in Th cells increased significantly(P<0.001,P<0.01);and the expression of CXCL9(P<0.001),CCL5(P<0.05),IFN-γ(P<0.01)and GZMB(P<0.05)increased in the lung tissue.In the lung metastasis model of B16 melanoma,PD could significantly reduce the number of lung tumor clones(P<0.01).Compared with the tumor model group,the proportion and number of CXCR3+ Th cells and IFN-γ+ Th cells in lung tissue of PD group increased(P<0.001,P<0.01),the transcription levels of chemokines CXCL9 and CCL5 were up-regulated in lung tumor microenvironment,and the transcription levels of IFN-γ and GZMB in lung Th1 cells were also significantly up-regulated.Conclusion:PD can enhance the expression of lung chemokines CXCL9 and CCL5,promote the recruitment of CXCR3+ Th1 cells in the lung tumor microenvironment,enhance the expression of local IFN-γ and GZMB,improve the lung immune microenvironment,and play an antagonistic pharmacological effect on lung metastasis.
