复方蟾酥胶囊对小鼠Lewis肺癌移植瘤的药效学研究
Efficacy of Compound Toad Venom Capsules on Xenograft Tumor in Lewis Lung Cancer Mice
摘要
Abstract
Objective:To investigate the inhibitory effect of different doses of Compound Toad Venom Capsules on Lewis lung cancer in C57BL/6J mice.Methods:Sixty SPF male C57BL/6J mice were selected and randomly divided into blank group(n = 10)and model group(n = 50).Blank group was raised separately.Mouse Lewis lung cancer cell line(LLC)was used for subcutaneous modeling of the right axilla of lung cancer tumor in the modeling group.After successful modeling,the group was randomly sub-divided into model group,cisplatin group and compound toad venom capsule low-dose,medium-dose and high-dose groups,with 10 mice in each group.The cisplatin group was intraperitoneally injected with cisplatin every other day,the low,medium and high dose groups of Compound Toad Venom Capsules were given different doses of compound toad venom capsule solution[0.1365,0.273,0.546 g/(kg·d)]every day,and the blank group was not treated.The model group was given physiological saline by gavage,and after 14 days,the mice were euthanized and tumor tissue and lung tissue were removed.The content of tumor necrosis fact-α(TNF-α),mRNA expression and protein expression of Caspase-3,vascular endothelial growth factor(VEGF)and lung pathology were detected by ELISA,real-time fluorescent quantitative PCR,Western blot and HE staining,to measure tumor suppression.Results:Compared with model group,the tumors in the medium dose group of compound toad venom capsule and the cisplatin group were significantly reduced(P<0.05),TNF alpha levels decreased(P<0.05,P<0.01),Caspase-3 mRNA and protein expression levels were up-regulated significantly(P<0.05,P<0.01,P<0.0001),VEGF mRNA and protein expression levels were significantly down-regulated(P<0.01,P<0.001).Alveolar wall thickening was observed in the model group,accompanied by more lymphocyte and neutrophil infiltration.Epithelial cell necrosis,nuclear fragmentation,pulmonary edema and hemorrhage were observed in local bronchus.Cisplatin group and compound toad venom capsule dosage groups can significantly improve the above conditions,and there is eosinophilic sero-like substance exudation in alveolar cavity.Conclusion:Compound toad venom capsule has the effect of inhibiting the growth of Lewis lung cancer tumor.关键词
