刺五加苷B影响EMT进程抑制肺癌细胞迁移和侵袭的机制研究
Eleutheroside B Inhibites Migration and Invasion of Lung Cancer Cell through EMT Progression

Abstract
Objective:To investigate the effects of Eleutheroside B on migration and invasion of human lung cancer cells A549 and H460.Methods:Lung cancer cells A549 and H460 were treated with different concentrations of Eleutheroside B(0,5,15,20,25,30,35,40,45 mmol/L),and control group and blank group were set up at the same time to find the optimal concentration.Lung cancer cells were treated with the optimal drug concentration for cell scratch test and Transwell migration and invasion test to observe the changes in cell scratch area and the number of cells passing through Transwell chamber and to explore the effect of Eleutheroside B on the migration and invasion of lung cancer cells A549 and H460.Subsequently,Western blot was used to detect the expression of epithelial mesenchymal transformation(EMT)related proteins E-cadherin,Slug and Snail and to explore the effect of Eleutheroside B on the EMT of lung cancer cells A549 and H460.Results:Eleutheroside B significantly inhibited the scratch area in lung cancer A549 and H460 cells,and significantly reduced the number of cells passing through the Transwell compartment and stromal gum.Compared with the control group,Eleutheroside B significantly up-regulated the expression of epithelial marker E-cadherin protein,and significantly reduced the expression levels of EMT transcription factors Snail and Slug.Conclusion:Eleutheroside B can significantly inhibit the growth of lung cancer cells A549 and H460,and inhibit cell migration and invasion by affecting the progression of EMT.关键词
