淫羊藿次苷Ⅱ改善APP/PS1双转基因小鼠认知功能的研究
Study on the improvement of cognitive function in APP/PS1 double-transgenic mice by Icariside Ⅱ

Abstract
Objective To study the improvement effects of Icariside Ⅱ(ICS Ⅱ)on cognitive function in APP/PS1 double-transgenic mice.Methods 30 7-month-old APP/PS1 mice were selected and randomly divided into the model control group(n=15)and ICS Ⅱ treatment group(n=15),and another 15 C57BL/6 mice of the same age were selected as the blank control group.ICS Ⅱ treatment group was gavaged with 10 mg·kg-1 of ICS Ⅱ,while the blank control group and the model control group were administrated with same amount of carboxymethyl cellulose sodium(CMC-Na)once a day for 28 days.On the 20th day,the ability of daily living of mice was tested by nest-building test,and the cognitive function of mice was tested by Morris water maze test.Hematoxylin-eosin(H & E)staining and Nissl staining were used to detect the pathological damage in hippocampal cortex,CA3 and DG regions.Immunofluorescence assay was carried out to detect the the deposition of β-amyloid protein in the cortex and hippocampus,as well as the activation of astrocytes and microglia of the mice.The content of malondialdehyde(MDA)and the activity of superoxide dismutase(SOD)in the brain of the mice were detected by TBA method and hydroxylamine method,respectively.Results Compared with the model control group,ICS Ⅱ could significantly improve the ability of daily living and cognitive function of APP/PS1 mice,alleviate the pathological damages of brain tissues,increase the number of neurons,reduce the deposition of Aβ-amyloid protein,inhibit the overactivation of astrocyte and microglia,increase the activity of SOD,and decrease the level of MDA in the brain.Conclusion ICS Ⅱ can improve the cognitive function of APP/PS1 double-transgenic mice by modulation of multiple pathological mechanisms.关键词
