牛至香酚对LPS免疫应激小鼠血清细胞因子含量和肠道细胞因子、紧密连接蛋白表达水平的影响
Effects of oregano phenol on serum cytokine content and mRNA expression of intestinal cytokines and tight junction proteins in mice with immune stress induced by LPS

Abstract
[Objective]The study aimed to explore whether oregano can effectively improve the immune function of mice under im-mune stress caused by lipopolysaccharide(LPS).[Method]Eighty mice were randomly divided into blank control group,model group,and test groups.The test groups were administrated with low(25 mg·kg-1),medium(50 mg·kg-1)or high(100 mg·kg-1)dose of oregano phenol while the blank control group and model group were administrated with 10 mL·kg-1 olive oil once a day for 14 d.At day 15,the test groups and model group were intraperitoneally injected with 8 mg·kg-1 LPS,and the blank group were injected with same amount of saline.Then blood and intestinal tissues of mice were collected 6 h later to determine the contents of serum cytokines(IL-1β,IL-6,TNF-α and IL-10)and mRNA expression levels of intestinal cytokines(IL-1β,IL-6,TNF-α and IL-10)and tight junction proteins(ZO-1,Occludin and Claudin-1).[Result](1)Compared with the blank control group,serum TNF-α(P<0.05),IL-10(P<0.05)and IL-1β(P<0.05)and IL-6(P<0.01)in the model group were significantly increased.The mRNA expression levels of TNF-α,IL-6 and IL-1β in the ileum and colon of the model group were significantly up-regulated(P<0.01),while that of ZO-1 were down-regulated(P<0.05);the mRNA expression levels of Occludin and Claudin-1 in the colon of the model group were down-regulated(P<0.01).(2)Compared with the model group,serum IL-6 and IL-1β were extremely signif-icantly(P<0.01)and significantly(P<0.05)decreased in all the test groups,serum TNF-α was only significantly decreased in the high-dose test group(P<0.01),and serum IL-10 was significantly increased(P<0.05)in both medium-and high-dose test groups.(3)Compared with the model group,the mRNA expression levels of IL-1β,TNF-α and IL-6 in the ileum of medium-and low-dose test groups were extremely significantly down-regulated(P<0.01),while those of ZO-1 and Occludin in the ileum of medium-and high-dose groups were up-regulated(P<0.01).Significantly increased mRNA expression levels of IL-10,Occludin and Claudin-1 only occurred in the colon of high-dose test group(P<0.01),while that of TNF-α was down-regulated(P<0.01).[Conclusion]O-regano phenol regulated the immune function of mice with LPS-induced immune stress by altering the serum cytokine content and ex-pression levels of intestinal cytokines and tight junction protein.关键词
