柯乐猪异常乳腺组织的TMT蛋白组学分析
Proteomic analysis of abnormal mammary gland tissue of Kele pig based on TMT technique

Abstract
[Objective]Explore the regulatory mechanisms of mammary gland development in Kele pigs using tandem mass tag(TMT)quantitative proteomics technology,providing a basis for breeding efforts.[Method]Three normal and three abnormal mam-mary gland tissues were collected from Kele sow pigs.TMT quantitative proteomics technology was employed to investigate the protein expression levels in normal and abnormal mammary gland tissues.Differential key proteins related to mammary gland development were screened,and bioinformatics analysis was conducted on the differentially expressed proteins.[Result]The results revealed 474 differentially expressed proteins,including 245 upregulated and 229 downregulated proteins.Gene ontology(GO)functional enrich-ment analysis indicated that these differentially expressed proteins were mainly enriched in biological processes such as epithelial cell differentiation,skin development,and renal tubule epithelial cell process.They were predominantly localized in extracellular regions and extracellular spaces,and involved in functions like peptidase inhibitor activity and peptidyl-cysteine activity.Kyoto encyclopedia of genes and genomes(KEGG)enrichment analysis revealed that these differentially expressed proteins were mainly enriched in sig-naling pathways related to complement system,peroxisome proliferator activated receptor,and Staphylococcus aureus infection.Dif-ferential protein interaction network analysis identified 10 key differentially expressed proteins,which included upregulated proteins of P14287(osteopontin),F1RXF9(keratin 25),I3LDS3(keratin 10),Q75QW1(epithelial cell adhesion molecule),A0A5S8KLN1(cornulin)and F1RW75(grifin),and downregulated proteins of A0A286ZT13(albumin),P06867(plasmino-gen),F1SCC9(protein containing serine protease inhibitor domain)and P50447(α-1-antitrypsin).[Conclusion]TMT quantitative proteomics was proved to be an effective technique in screening differentially expressed proteins between normal and abnormal mam-mary gland tissues of sows.关键词
