MiR-184 promotes the proliferation ability of ovarian cancer cells by regulating FOXO3

Abstract
Objective To investigate the effect of miR-184 on the proliferation of ovarian cancer cells by regula-ting FOXO3.Methods The expression levels of miR-184 in samples were detected using fluorescence quantitative PCR.miR-184 mimic and miR-184 inhibitor were transfected into human SKOV3 ovarian cancer cells.The MTT assay was used to detect the in vitro proliferation activity of successfully transfected human SKOV3 ovarian cancer cells.The ex-pression levels of cyclin D1,p27,and FOXO3 mRNA were also measured.Results The expression of miR-184 in ovar-ian cancer tissues and cells was significantly higher than that in adjacent tissues and human ovarian epithelial cells IOSE80.Compared to IOSE80 cells,SKOV3 cells showed a significant decrease in FOXO3 and p27 mRNA expression levels,and a significant increase in cyclin D1 mRNA expression levels.The MTT assay indicated that overexpression of miR-184 significantly increased the proliferation activity of SKOV3 cells,while inhibition of miR-184 expression signif-icantly decreased their proliferation activity.Furthermore,overexpression of miR-184 resulted in a significant increase in cyclin D1 mRNA expression and a significant decrease in FOXO3 and p27 mRNA expression in SKOV3 cells(P<0.05).Conversely,inhibition of miR-184 expression led to a significant decrease in cyclin D1 mRNA expression and a signifi-cant increase in FOXO3 and p27 mRNA expression in SKOV3 cells(P<0.05).Conclusion MiR-184 can affect the proliferation capability of ovarian cancer cells and regulate the expression of cell cycle-related genes.关键词
