外周血miRNA差异性表达与系统性红斑狼疮病情及预后的关系
Differential expression of peripheral blood miRNA and their relationship with disease severity and prognosis in systemic lupus erythematosus

Abstract
Objective To investigate the relationship between differential expression of peripheral blood miRNA and disease severity and prognosis in patients with systemic lupus erythematosus(SLE).Methods Patients with SLE from the Department of Rheumatology and Immunology between October 2020 and October 2022 were selected.Next-gen-eration sequencing(NGS)was performed to assess the miRNA content in peripheral blood mononuclear cells(PBMC)from 4 SLE cases and 4 healthy controls.Quantitative polymerase chain reaction(qPCR)was conducted in 32 SLE cases and 32 healthy controls to validate candidate miRNA.Additionally,receiver operating characteristic(ROC)curve analy-sis was carried out to evaluate diagnostic performance.Results Compared to healthy controls,SLE cases exhibited signif-icant upregulation of 157 miRNA and downregulation of 110 miRNA in PBMC.Notably,miR-183-5p was increased,and miR-374b-3p was decreased,with statistically significant differences(P＜0.05).The expression level of miR-183-5p was positively correlated with the Systemic Lupus Erythematosus Disease Activity Index(SLEDAI)and anti-ds-DNA antibody levels.Conclusion This study reveals a significant alteration in the miRNA expression profile in PBMC of SLE cases compared to healthy controls.MiR-183-5p emerges as a potential diagnostic biomarker for SLE,and its ex-pression is associated with disease activity in SLE.关键词
