为探索适合不同类型竹种竹叶总RNA的提取方法,本研究选取合轴丛生型竹种勃氏甜龙竹、合轴散生型竹种云南箭竹、单轴散生型竹种毛竹、复轴混生型竹种巴山木竹为代表,提取其心叶的总RNA,通过琼脂糖凝胶电泳检测RNA的完整性,利用超微量紫外分光系统检测RNA的纯度和浓度,比较自配TRIzol试剂法、商品化TRIzon试剂法、RNA提取试剂盒法、改良十六烷基三甲基溴化铵(CTAB)法四种方法提取的竹叶总RNA的质量、纯度和提取成本的差异.结果表明,自配TRIzol试剂法可以提取出质量好、浓度高的竹叶总RNA,且提取成本较低;商品化TRIzon试剂法提取的总RNA质量及浓度仅次于自配TRIzol试剂法,提取成本稍高;而RNA提取试剂盒法和改良CTAB法无法提取出高浓度的竹叶总RNA.本研究结果为竹类植物更深入的分子生物学研究提供了一定的基础,并为其他竹类植物总RNA的提取提供了参考依据.

To investigate the total RNA extraction method from various types bamboo,we selected the core leaves of Dendrocalamus brandisii(sympodial rhizomes with caespitose culms),Fargesia yunnanensis(sympodial rhizomes with scattered culms),Phyllostachys edulis(monopodial rhizomes),and Bashania fargesii(amphipodial rhizomes)as the experimental materials for total RNA extraction.The integrity of RNA was assessed using agarose gel electrophoresis,while the purity and concentration of RNA were determined using ultra-micro ultraviolet Photosystem.A comparison was made among the self-made TRIzol reagent,TRIzon reagent,RNA extraction kit,and improved CTAB methods regarding the quality,purity of the extracted total RNA,and the cost.We found that with self-made TRIzol reagent method,the quality of total RNA was superior and the concentration was higher but with a comparatively lower cost.With the TRIzon reagent method resulted in total RNA with slightly lower quality and concentration,albeit with a slightly higher extraction cost.However,both the RNA extraction kit and the improved CTAB method were found to be ineffective in extracting total RNA of high concentration and good quality from bamboo leaves.The findings of this study serve as a foundation for future research in the field of molecular biology pertaining to bamboo plants,and also offer valuable insights for the extraction of total RNA from other bamboo species.
