为探究抑制果实浮皮的方法及赤霉素(GA3)与二氢茉莉酸丙酯(PDJ)抑制浮皮发生的机制,本研究以完熟栽培温州蜜柑宫川为试验材料,通过喷施钙或GA3与PDJ,研究其对果实浮皮指数以及果实品质的影响,并分析GA3与PDJ处理后果皮的细胞壁结构、细胞壁物质含量、相关水解酶活性、果皮激素含量及相关基因表达量的变化.结果表明,各处理均能降低果实浮皮指数,其中采前喷施2次GA3和PDJ后果实浮皮指数始终维持在较低水平,且该处理条件下果实外观及内在品质无明显变化.同时,GA3与PDJ处理下调了CitCx、CitPME等基因表达量,降低了果胶酯酶(PME)和纤维素酶(Cx)的活性,提高了果皮中原果胶和半纤维素含量,延缓了纤维素含量下降并增加了果实硬度,并且降低了果皮细胞膜脂过氧化程度.喷施GA3与PDJ后果皮中乙烯合成量下降而生长素含量上升,CitACO基因表达下调,而CitETR1及CitIAA27表达上调.综上,采前喷施2次GA3与PDJ是防治完熟栽培温州蜜柑浮皮的最佳方法.GA3与PDJ通过抑制细胞壁结构物质降解,降低膜脂过氧化程度,同时抑制乙烯合成并降低果皮细胞对乙烯的敏感性,提高果皮生长素(IAA)含量并抑制果皮衰老,从而抑制浮皮的发生.本研究结果为开发控制浮皮技术及揭示柑橘果实浮皮发生机制提供了实践与理论参考.

In order to investigate the methods of preventing citrus fruit from puffing and the mechanism of gibberellin(GA3)and propyldihydrojasmon(PDJ)inhibiting peel puffing,the effects of calcium or GA3 and PDJ on the fruit puffing index and fruit quality were examined by foliar spraying on satsuma mandarin Miyagawa with delayed harvest cultivation,and the alterations in peel cell wall structure,cell wall substance content,hydrolytic enzyme activity,phytohormone content,and the relevant gene expression with GA3 and PDJ-treated were analyzed.The results demonstrated that all treatments were able to reduce the fruit puffiness index.However,only spraying twice with GA3 and PDJ-treat before harvest could control the fruit puffiness index at a low level without affecting the external and internal quality of the fruit.In the GA3-and-PDJ treatment,the activity of pectin methylesterase(PME),cellulase(Cx)and the expression of CitPME and CitCx genes was suppressed.The protopectin and hemicellulose content in the peel and fruit hardness were increased in GA3-and-PDJ treatment.Compared with congtrol,cellulose content decreasing was delayed,and the degree of membrane lipid peroxidation in the peel was reduced.Meanwhile,the expression of the CitACO gene was suppressed,which resulted in a decrease of ethylene synthesis,while the CitETR1 gene was up-regulated,and the IAA content and CitIAA27 gene expression increased.The results of this study indicated that combined spraying of GA3-and-PDJ twice was the most effective method to prevent the peel puffing of satsuma mandarin with delayed harvest cultivation.The treatment inhibited the degradation of cell wall structural substances,reducing the degree of membrane lipid peroxidation,decreasing ethylene synthesis and reducing the sensitivity of peel cells to ethylene,and improving peel IAA content,and thus inhibiting peel senescence and the occurrence of peel puffing.This research provides a theoretical and practical foundation for understanding the mechanism of citrus fruit puffing and developing strategies to control citrus puffing.
