红景天苷对Lewis肺癌小鼠恶性胸腔积液生成的影响
Effect of salidroside on the formation of malignant pleural effusion in mice with Lewis lung cancer

Abstract
Objiective To explore the inhibitory effect of salidroside(Sal)on malignant pleural effusion(MPE)of Lewis lung cancer metastases in mice.Methods An MPE model of C57BL/6 mice was established by thoracic injection of Lewis lung cancer cells.After modeling,they were randomly divided into a normal group,a model group,a Sal group,a cisplatin group(DDP group),and a salidroside+cisplatin group(Sal+DDP group).At the end of the experiment,the pleural fluid volume,the number and quality of metastatic tumors in chest wall were collected,and the survival cycle of mice in each group was recorded.The serum levels of tumor necrosis factor-α(TNF-a),hypoxia inducible factor-1α(HIF-1α)were detected by enzyme linked immunosorbent assay(ELISA).Real-time reverse transcription-PCR(Realtime RT-PCR)and western blot(WB)were used to detect the vascular endothelial growth(VEGF)and VEGF mRNA levels in the pleural fluid of mice.Results Compared with the model group,the survival cycle of MPE mice in the Sal+DDP combined group was significantly prolonged after treatment(P=0.0021).The volume of MPE,the number and quality of chest wall metastases in the Sal,DDP and combined groups were significantly reduced(P＜0.01).The level of TNF-α in peripheral blood of the Sal group,the DDP group and the combined group was significantly increased,while the level of HIF1α was significantly decreased(P＜0.05).After treatment,compared with the model group,the levels of VEGF and VEGF mRNA in pleural effusion in the Sal group,the DDP group and the combined group were decreased,and the difference was statistically significant(P＜0.05).Conclusion Salidroside combined with cisplatin in the treatment of MPE has a significant effect,which may be related to inhibiting HIF-1α-VEGF molecular pathway and activating TNF-α.关键词
