Motherwort Injection Promotes Angiogenesis Via Alkaloid-Induced Upregulation of Tie-2,VEGFR2 and VEGF in Zebrafish Modle

摘要

Abstract

Objective:To study the angiogenic effects of motherwort injection in a zebrafish vascular injury model,and to clarify the potential material basis and molecular mechanism.Methods:Using HPLC-PAD and TLC techniques to detect the main components of motherwort injection.Using sunitinib induced trans-genic Tg(flk 1:eGFP)zebrafish vascular injury model,after intervention with motherwort injection,the length of intersegmental blood vessels(ISVs)of zebrafish was measured,and the expression level of angio-genesis-related genes was detected by QT-PCR.Results:It was found that the main components of Leonu-rus heterophyllus injection were stachydrine,choline and trigonelline.Among them,motherwort injection,trigonelline and choline can alleviate chemically induced zebrafish vascular damage,and its mechanism in-volves that motherwort injection upregulates the expressions of Tie-2,VEGF and VEGFR2,and its main component trigonelline up-regulates the expressions of Tie-2 and VEGF,and choline up-regulates the ex-pressions of VEGF and VEGFR2.Conclusion:Motherwort injection and its main alkaloid components can promote angiogenesis by up-regulating Tie-2,VEGFR2 and VEGF.
