利用响应面法优化狐臭柴叶多糖提取工艺、单糖组成及生物活性研究
Optimization of Extraction Process,Monosaccharide Composition,and Biological Activity of Polysaccharides from Premna Puberula Pamp on Response Surface Methodology

Abstract
The extraction process of acid-soluble polysaccharide(PSAP)was optimized,and the monosaccharide composition and in vitro biological activity of PSAP were investigated.The leaves of Premna Puberula Pamp were used as raw materials.After drying,crushing,degreasing and decolorization,the extraction process of PSAP was opti-mized by response surface methodology on the basis of single factor.The composition and content of monosaccharides were determined by high performance liquid chromatography(HPLC).The biological activity in vitro of PSAP was studied by in vitro antioxidant activity test and tumor cell test.The results showed that the optimum extraction pro-cess of PSAP was as follows:extraction temperature was 100℃,liquid-solid ratio was 1∶31 g·mL-1,extraction time was 240 min,and pH value of oxalic acid-disodium hydrogen phosphate was 2.Under these conditions,the extrac-tion rate of PSAP was(31.04±0.21)%.PSAP was mainly composed of eight monosaccharides,and the peak area ac-counted for 0.22%of mannose,6.13%of rhamnose,4.58%of glucuronic acid,31.98%of galacturonic acid,18.31%of glucose,9.90%of galactose,10.23%of arabinose and 4.09%of fucose,respectively.At the same time,PSAP has strong antioxidant activity in vitro and certain reducing ability to iron ions.It has strong scavenging ability to 1,1-di-phenyl-2-picrylhydrazyl(DPPH)free radical,hydroxyl radical and 2,2'-azino-3-ethylbenzothiazoline-6-sulfonic acid(ABTS)free radical.The IC50 values were 3.69 mg·mL-1,2.91 mg·mL-1 and 0.43 mg·mL-1,respectively.Cell experiments showed that PSAP had a significant inhibitory effect on the proliferation and migration of A549 cells and HepG2 cells in a dose-dependent manner.The results of this study provide theoretical support for the extraction pro-cess,food development and clinical application of polysaccharides from Clerodendrum odoratum.关键词
