解毒活血汤对慢性肾脏病大鼠肾脏FGF23、Klotho表达的影响OACSTPCD
Effects of Jiedu Huoxue decoction on the expression of FGF23 and Klotho in the kidneys of rats with chronic kidney disease
目的:观察解毒活血汤对慢性肾脏病(CKD)大鼠肾脏FGF23、Klotho的影响.方法:将 60 只SPF级健康雄性 SD大鼠,随机分空白组、模型组、中药高剂量组、中药中剂量组、中药低剂量组,每组 12 只.采用腺嘌呤加高磷饮食制作CKD大鼠伴血管钙化模型.采用生化试剂盒测定血清中肌酐、尿素氮、钙、磷水平;Western blot 法检测肾脏组织中 FGF23、Klotho的表达.结果:与空白组相比,其余各组血清中肌酐、尿素氮、钙、磷均有明显变化(P<0.05 或P<0.01);和模型组相比,中药高剂量组、中药中剂量组、中药低剂量组 CKD 大鼠血清中肌酐、尿素氮、磷水平明显改善(P<0.05),FGF23、Klotho蛋白表达量增高(P<0.05),血清钙水平升高(P<0.05).结论:解毒活血汤可以改善CKD 大鼠肾脏功能,可以通过提高 CKD大鼠肾脏组织中 FGF23、Klotho 蛋白的表达影响失衡的 FGF23-Klotho轴,进而调节CKD大鼠血中钙、磷浓度.
Objective:To observe the effects of Jiedu Huoxue decoction on FGF23 and Klotho in rats with chron-ic kidney disease(CKD).Methods:Sixty healthy male SD rats with SPF grade were randomly divided into black group A,group B,group I,group H,and group G,with 12 rats in each group.A model of vascular calcification in rats with CKD was prepared using adenine plus high-phosphorus diet.Biochemical kits were used to determine Scr,BUN,Ca,P levels.The Western blot method detected the expression of FGF23,Klotho in kidney tissue.Results:Compared with the blank group,the serum serum Scr,BUN,Ca and P were significantly changed in the rest groups(P<0.05 or P<0.01).In compared to the model group,the serum levels of Scr,BUN and P in group G,group H and group I were significantly improved(P<0.05),the expression of FGF23 and Klotho proteins was increased(P<0.05),and the serum Ca level was increased(P<0.05).Conclusion:Jiedu Huoxue decoction can improve the kidney function of CKD rats,and can affect the unbalanced FGF23-Klotho axis by increasing the expression of FGF23 and Klotho pro-teins in the kidney tissues of CKD rats,and then regulate the concentration of Ca and P in the blood of CKD rats.
