养阴凉血方对过敏性紫癜患儿血清诱导内皮细胞损伤的保护作用
Protective effect of Yangyin Liangxue recipe on endothelial cells induced by serum in children with Henoch-schnlein purpura

Abstract
Objective:To investigate the protective effect and mechanism of Yangyin Liangxue recipe on endo-thelial cells induced by serum of children with Henoch-schonlein purpura(HSP).Methods:HUVECs were used as the research object,the serum of healthy children and children with HSP were to intervene,and different concentra-tions of Yangyin Liangxue recipe were added.They were divided into blank control group,normal control group,HSP group and Yangyin Liangxue recipe(0.5,1 g/ml)group.The supernatant was collected after 24 hours of culture.Cell morphology was observed under inverted microscope.The levels of TNF-α,IL-8,IgA,CD89 in the supernatant of each group were determined by ELISA.Western blot was used to detect the expression of PI3 K protein in each group.Results:The levels of TNF-α,IL-8,IgA,CD89 and the expression PI3 K protein in the supernatant of each group were significantly higher in HSP group than in the blank and normal control group(all P<0.05),and the number of cells was decreased and the morphology was shrunk.Compared with the HSP group,the levels of TNF-α,IL-8,IgA,CD89 and PI3K protein expression in the Yangyin Liangxue recipe two groups were significantly decreased(all P<0.05),and the trend of cell number reduction and morphological shrinkage was improved.Conclusion:Yangyin Liangxue recipe can treat Henoch-schonlein purpura and its mechanism may be related to regulating the secretion of cytokines such as TNF-α,IL-8,IgA and CD89 and reducing the phosphorylation level of PI3K protein in PI3K/AKT/NF-κB signaling pathway.关键词
