目的:研究蒙药蓝刺头对去卵巢大鼠骨代谢标志物P1 NP、β-CTX的影响.方法:将 72 只 4 月龄 SD系清洁级雌性大鼠随机分为六组,每组 12 只,分别为假手术组、去卵巢骨质疏松模型组、蓝刺头高剂量组、中剂量组、低剂量组及淫羊藿组.除假手术组外,其余各组大鼠均采用卵巢摘除去势法建立绝经后骨质疏松模型.术后正常饲养 90 d,假手术组和模型组大鼠灌胃等体积 0.9%氯化钠溶液,各治疗组大鼠灌胃相应药物,每天 1 次,连续 90 d.末次给药 24h后,处死大鼠,采用 Micro-CT 测定大鼠股骨骨密度,酶联免疫法检测大鼠血清中 P1 NP、β-CTX、TNF-α、IL-1β、IL-6 的含量.结果:与假手术组比较,模型组大鼠股骨近端 BMD 明显降低,血清中 P1 NP、β-CTX、TNF-α及 IL-1β和 IL-6 的浓度明显升高(P<0.05).与模型组比较,蓝刺头组的BMD升高,血清中 P1 NP、β-CTX、TNF-α及 IL-1β和 IL-6 的浓度下降(P<0.05).结论:蒙药蓝刺头可降低去卵巢大鼠的P1NP和β-CTX的含量,同时减少TNF-α、IL-1β、IL-6 的浓度,进而达到增加骨含量,减少骨吸收的作用.

Objective:To study the effects of Eechinops latifolius on bone metabolic markers P1 NP and β-CTX in ovariectomized rats.Methods:Seventy-two 4-month-old SD SPF grade female rats were randomly divided into sham group,ovariectomized osteoporosis model group,Mongolian medicine Cyanostis high-dose group,medium-dose group,low-dose group and Epimedium group,12 rats in each group.Except sham operation group,the other groups were treated with ovary removal and castration to establish postmenopausal osteoporosis model.Rats in sham opera-tion group and model group were fed normal saline for 90 days after operation,and rats in each treatment group were intragastric with corresponding drugs once a day for 90 days.24 h after the last administration,the rats were killed,and the femoral bone mineral density of the rats was determined by Micro-CT,and the serum levels of P1 NP,β-CTX,TNF-α,IL-1βand IL-6 were detected by enzyme-linked immunoassay.Results:Compared with sham group,the proxi-mal femur BMD of OVX group was significantly decreased,and the serum contents of P1 NP,β-CTX,TNF-α,IL-1β and IL-6 were significantly increased(P<0.05).Compared with OVX group,BMD was increased and serum con-tents of P1 NP,β-CTX,TNF-α,IL-1βand IL-6 were decreased in treatment group(P<0.05).Conclusion:Mongolian medicine Lanacitou can decrease the contents of P1 NP andβ-CTX in ovariectomized rats,and at the same time de-crease the concentrations of TNF-α,IL-1βand IL-6,thereby increasing bone content and decreasing bone absorption.
