夏桑菊浸膏对高渗诱导干眼模型人角膜上皮细胞保护机制研究

Abstract
Objective To investigate the protective effect and mechanism of the extract of Xiasangju on human corneal epithelial cells induced by hypertonic.Methods Hypertonic model was constructed with NaCl,and cell viability was detec-ted by MTS method.The expressions of IL-1β,IL-6 and IL-18 were detected by RT-qPCR and enzyme-linked immu-nosorbent assay.Intracellular reactive oxygen species(ROS)were detected by 2',7'-dichlorofluorescein yellow diacetate fluorescence probe.Results Compared with the control group,the cell vitality of the model group at 600 mOsm·L-1 was sig-nificantly decreased,and the cell vitality of the model group was significantly increased after treatment with the extract of Xi-asangju.In terms of inflammatory indicators,the secretion levels of IL-1β,IL-6 and IL-18 and the expression levels of IL-1β in 450 mOsm·L-1 cells in the model group were significantly increased compared with the control group.Compared with 450 mOsm·L-1 model group,the above indexes were significantly decreased after treatment with the extract of Xiasangju.In the antioxidant study,the reactive oxygen species in 450 mOsm·L-1 cells in the model group was significantly increased,while the reactive oxygen species was significantly decreased after treatment with Xiasangju extract.Conclusion The extract of Xiasangju can inhibit the decrease of human corneal epithelial cell vitality induced by hypertonic,and can protect human corneal epithelial cells in hypertonic environment by inhibiting the production of inflammatory factors and inhibiting the increase of reactive oxygen species.关键词
