脑胶质瘤中PROX1表达与患者预后的关系及其生物学作用
Relationship between PROX1 expression and prognosis in brain glioma patients and its biological role

Abstract
Objective To investigate the relationship between the prospero-related homeobox 1(PROX1)expression and the prognosis of patients with brain glioma and its biological role.Methods The expression level of PROX1 and the survival time in brain glioma patients were analyzed using gene expression profiling interactive analysis(GEPIA)database.A total of 75 patients with brain glioma admitted to the department of neurosurgery in the First Affiliated Hospital of Air Force Medical University of PLA from May 2015 to December 2021 were selected.And the tumor tissue samples and adjacent normal tissue samples were collected,respectively.PROX1 expression level in tissues was detected by immunohistochemical staining,and the survival of patients was analyzed according to follow-up data.U251 cells were divided into control group,si-NC group and si-PROX1 group.U251 cells in si-NC group and si-PROX1 group were transfected with si-NC and si-PROX1 respectively,while U251 cells in control group were not transfected.The proliferation level of U251 cells was detected by EdU.The apoptosis of U251 cells was detected by flow cytometry.The migration and invasion abilities of U251 cells were detected by Transwell.The mRNA and protein expression levels of PROX1,VEGFC and VEGFR3 in U251 cells were detected by qRT-PCR and Western blot.Results The results of GEPIA database analysis showed that PROX1 was highly expressed in glioblastoma tissues.The results of clinical tissue sample analysis showed that PROX1 was highly expressed in brain glioma tissues,and the survival time of patients with high PROX1 expression was lower than that of patients with low PROX1 expression(P<0.01).Compared with control group and si-NC group,the mRNA and protein expression levels of PROX1 in U251 cells in si-PROX1 group were decreased(P<0.01).Compared with control group and si-NC group,the proliferation of U251 cells was decreased in si-PROX1 group(P<0.01),and the apoptosis was increased(P<0.01),the numbers of migratory and invasive U251 cells were decreased(P<0.05).Compared with control group and si-NC group,the mRNA and protein expression levels of PROX1,VEGFC and VEGFR3 in U251 cells in si-PROXl group were decreased(P<0.05).Conclusion The high expression level of PROX1 is a negative factor for the prognosis of patients with brain glioma.PROX1 can inhibit the apoptosis of GBM cells and promote the prolifera-tion,migration and invasion of GBM cells,thus promoting the malignant development of GBM.关键词
