黄芪多糖通过调控JAK2/STAT3信号通路抑制膀胱癌UM-UC-3细胞增殖、迁移与侵袭
Astragalus polysaccharide inhibits proliferation,migration and invasion of bladder cancer UM-UC-3 cells by regulating JAK2/STAT3 signaling pathway

Abstract
Objective To investigate the effect and its mechanism of Astragalus polysaccharide(APS)on the proliferation,migration and invasion abilities of bladder cancer UM-UC-3 cells.Methods Bladder cancer UM-UC-3 cells were treated with 0.1%DMSO and different concentrations of APS(250,500,1 000 mg/L)for 24,48,72 h,respectively,and then,the proliferation ability of UM-UC-3 cells was detected by CCK-8 assay.At 24 h,the cell migration and invasion abilities were detected using wound healing assay and Transwell invasion assay,the cell cycle of UM-UC-3 cells was detected by flow cytometry,and the expressions of JAK2,p-JAK2,STAT3 and p-STAT3 proteins were detected by Western blot.Results Compared with DMSO group,the proliferation ability,the wound healing rate and the invasion ability of UM-UC-3 cells were significantly reduced in 250,500,1 000 mg/L APS groups(P<0.05).The proportion of UM-UC-3 cells in G0/G1 phase in APS groups were significantly increased compared with DMSO group(P<0.05).The protein expressions of p-JAK2 and p-STAT3 in 500 and 1 000 mg/L APS groups were decreased compared with DMSO group(P<0.05),and the protein expressions gradually decreased with the increase of APS concentration(P<0.05).Conclusion APS can inhibit the proliferation,migration and invasion abilities of bladder cancer UM-UC-3 cells,and block the cell cycle in GO/G1 phase by inhibiting the activation of JAK2/STAT3 signaling pathway.关键词
