Effect of SUSD2 gene on proliferation and invasion of ovarian cancer cells and its mechanism

Abstract
Objective To investigate the effect of sushi domain containing 2(SUSD2)gene on the proliferation and invasion of ovarian cancer cells and its mechanism.Methods The SUSD2 gene expression in ovarian cancer and normal ovarian tissue and its effect on the survival rate were analyzed based on GEPIA online tool.SUSD2 gene expression level was detected in 60 cases of ovarian cancer tissues and 30 cases of normal ovarian tissues from department of pathology in our hospital by immunohistochemical SP method.SKOV-3 cells were transfected with lentiviral vectors containing SUSD2 gene interference fragments or meaningless fragments,and stable expression strains were screened and named as sh-SUSD2 group and sh-NC group respectively.Cell proliferation was evaluated by CCK-8 and plate clonogenic assay,and cell migration and invasion were evaluated by Transwell assay.The SUSD2 coexpression gene set was screened through online tools,and then the highly related gene TRIM29 was selected as the coexpression gene of SUSD2.The TRIM29 expression in ovarian cancer and normal ovarian tissues were analyzed based on GEPIA online tool,then the difference of TRIM29 in pathological specimens from our hospital was analyzed by immunohistochemical SP method.The expression levels of SUSD2 and TRIM29 in SKOV-3 cells were detected in sh-SUSD2 group and sh-NC group by Western blot.Results The results of GEPIA analysis and SP immunohistochemistry showed that SUSD2 gene expression in ovarian cancer was significantly higher than that in normal tissues(P<0.05).The overall survival rate of ovarian cancer patients with high SUSD2 gene expression was lower than those with low SUSD2 gene expression(P<0.05).Compared with sh-NC group,the proliferation in sh-SUSD2 group was significantly inhibited by CCK-8 and clonogenic assay(P<0.05).Compared with sh-NC group,the migration and invasion abilities of SKOV-3 cells in sh-SUSD2 group were significantly decreased(P<0.05).Online tool analysis showed that SUSD2 was positively correlated with TRIM29(Spearman r=0.46,Pearson r=0.47).The expression of TRIM29 in ovarian cancer was significantly higher than that in normal ovarian tissue(P<0.05).The expression of TRIM29 in sh-SUSD2 group was significantly lower than that in sh-NC group by Western blot(P<0.05).Conclusion SUSD2 gene promotes the proliferation and invasion of ovarian cancer cells,and it is closely related to the poor prognosis of ovarian cancer patients.SUSD2 gene may promote the development of ovarian cancer through TRIM29 pathway.关键词
