Effects of EIF4G1 on proliferation,invasion,migration,and apoptosis of endometrial cancer cells

Abstract
Objective To investigate the expression of eukaryotic translation initiation factor 4G1(EIF4G1)in endometrial carcinoma(EC)tissue and its effects on the proliferation,invasion,migration,and apoptosis of EC cells.Methods Firstly,real-time quanti-tative PCR and immunohistochemistry were used to detect the relative expression of EIF4G1 mRNA and the positive protein expression rate in tumor tissues and corresponding normal tissues adjacent to cancer in 20 patients with EC.Then,the EIF4G1 overexpression plasmid and its control plasmid(pCMV6),and the low-expression plasmid and its control plasmid(PLKO.1)were constructed in vitro,and then transfected into human EC cell line Ishikawa using LipofectamineTM2000 transfection reagent.Western blot was used to screen the cell lines stably expressing EIF4G1 protein.The experiment was divided into five groups:blank control group,overexpres-sion group,overexpression control group,low-expression group,and low-expression control group.MTT was used to detect the cell proliferation,Transwell method was used to detect the cell invasion,the scratch test was used to detect the cell migration,and TUNEL method was used to detect the apoptosis rate.Results The relative expression of EIF4G1 mRNA and the positive protein expression rate were significantly higher in tumor tissues than those in normal tissues adjacent to cancer(P<0.05).Compared with blank control group,EIF4G1 protein was significantly increased in overexpression group but significantly decreased in low-expression group(P<0.05).Compared with blank control group and overexpression control group,the cell proliferation rate in overexpression group was significantly increased(P<0.05),and the number of cell invasion and the migration distance were increased(P<0.05),while the apoptosis rate was decreased(P<0.05).Compared with blank control group and low-expression control group,the proliferation rate in low-expression group was significantly decreased(P<0.05),and the cell invasion number and the migration distance were also decreased(P<0.05),while the apoptosis rate was increased(P<0.05).Conclusion EIF4G1 may be an oncogene to participate in the occurrence of EC and regulate the malignant biological behaviors of cells.EIF4G1 is expected to become a targeted intervention site for EC.关键词
