MEHP对正常肝细胞脂肪酸分解代谢相关基因的影响
Effect of MEHP on genes involved in fatty acid catabolism in normal hypatocytes

Abstract
Objective To investigate the effects of mono-(2-ethylhexyl)phthalate(MEHP)on fatty acid catabolism-related genes in HL-7702 hepatocytes.Methods HL-7702 cells were exposed to different concentrations of MEHP(0,0.01,1,10,100 μmol/L)and 1 mmol/L oleic acid(OA)for 24 h and 48 h,respectively.The cells without exposure to MEHP were used as negative control group.The viability of cells was detected by MTT assay,and qRT-PCR was used to detect the transcriptional levels of miRNA-34a and genes(SIRT1,PPARα,CPT1A)related to fatty acid catabolism in cells after exposure.Results Compared with negative control group,the survival rate of cells in OA group decreased after 24 h of exposure(P<0.05),and the survival rate of cells in various concentrations of MEHP groups did not change significantly(P>0.05).Compared with negative control group,the expression levels of miRNA-34a in 10,100 μmol/L MEHP groups and OA group significantly decreased after 24 h of exposure(P<0.05).Compared with negative control group,the miRNA-34a expression level in 0.01 μmol/L MEHP group significantly increased after 48 h of exposure(P<0.05).Compared with negative control group,the mRNA expression levels of SIRT1,PPARα and CPT1A in 0.01,1,10,100 μmol/L MEHP group and OA group showed no significant changes.Conclusion Different concentrations of MEHP have no effect on the cellular viability of hepatocytes.MEHP can reduce the expression level of miRNA-34a,but it does not change the mRNA levels of fatty acid catabolism-related genes.关键词
