槐杞黄对高糖诱导HK-2细胞损伤的保护效应及机制
Therapeutic effect and mechanism of Huaiqihuang on high glucose-induced HK-2 cell damage

Abstract
Objective To investigate whether Huaiqihuang(HQH)inhibits oxidative stress and apoptosis in high glucose-induced human renal tubular epithelial cells(HK-2 cells)by modulating the AKT/GSK3β signaling pathway.Methods HK-2 cells were divided into normal control group,mannitol group,high-glucose model group,Huaiqihuang group,and captopril group.CCK-8 method was used to detect HK-2 cell viability,reactive oxygen species(ROS)assay kit was used to detect ROS content,malondialdehyde(MDA)assay kit was used to detect MDA content,superoxide dismutase(SOD)activity assay kit was used to detect SOD content,RT-qPCR was used to detect kidney injury molecule-1(KIM-1)and neutrophil gelatinase-associated lipocalin(NGAL)mRNA expres-sion levels,and Western blot was used to detect the expression levels of apoptosis-related proteins(Bax,Bcl-2,cleaved-Caspase-3,Caspase-3)and AKT/GSK3β pathway proteins(p-AKT,AKT,p-GSK-3β,GSK-3β).Results Compared with normal control group,HK-2 cell viability was significantly decreased in high-glucose model group(P<0.001),KIM-1 and NGAL mRNA expression levels were significantly increased(P<0.001),ROS level was increased(P<0.001),MDA content was significantly elevated(P<0.001),SOD activity was significantly decreased(P<0.001),Bcl-2 protein level was significantly downregulated(P<0.001),Bax and cleaved-Caspase-3 protein levels were significantly upregulated(P<0.001),p-AKT protein level was significantly downregulated(P<0.001),and p-GSK-3β protein level was significantly upregulated(P<0.001).Compared with high-glucose model group,HK-2 cell viability was significantly increased in Huaiqihuang group and captopril group(P<0.001),KIM-1 and NGAL mRNA expression levels were significantly decreased(P<0.001),ROS level was reduced(P<0.001),MDA content was significantly decreased(P<0.001),SOD activity was significantly increased(P<0.001),Bcl-2 protein level was significantly upregulated(P<0.01),Bax and cleaved-Caspase-3 protein levels were significantly downregulated(P<0.01),p-AKT protein level was significantly upregulated(P<0.01),and p-GSK-3β protein level was significantly downregulated(P<0.01).All those indexes showed no significant difference between Huaiqihuang group and captopril group.Conclusion Huaiqihuang exhibits inhibitory effects on oxidative stress and apoptosis in high glucose-induced HK-2 cells,which may be related to the modulation of the AKT/GSK3β signaling pathway.关键词
