颐神解郁针刺法对抑郁症大鼠的治疗作用及其机制
Therapeutic effect and its mechanism of Yishen Jieyu acupuncture on depression rats

Abstract
Objective To observe the effects of Yishen Jieyu acupuncture combined with electroacupuncture on the behavior and the pyroptosis-related proteins in hippocampal tissue of depression rats,and explore its mechanism in the treatment of depression.Methods Twenty-eight healthy male Wistar rats were randomly divided into normal group,model group,electroacupuncture group and sham acupuncture group,with 7 rats in each group.The model was established by solitary rearing combined with chronic unpredictable mild stress(CUMS).Rats in electroacupuncture group were subjected to a density wave in acupoints Baihui and bilateral Ganshu with a frequency of 2 Hz and an intensity of periacupoint muscle microfibrillation which did not cause rat struggle or hiss.The rats were intervened for 20 min once a day,and the intervention lasted 6 d every week for 4 weeks.The acupoints in sham acupuncture group were selected 5 mm away from the standard acupoint level,and the other criteria were consistent with those in electroacupuncture group.The rats in normal group and model group were not given any intervention.The body weight of rats was observed and recorded,and the sucrose preference test and the open field test were performed to evaluate the behaviors.After that,the contents of NOD-like receptor thermal protein domain associated protein 3(NLRP3)and Gasdermin D(GSDMD)in rat hippocampus were detected by Western blotting.Results Compared with normal group,the body weight,the sucrose preference rate,the horizontal exercise score and the vertical exercise score in model group decreased(P＜0.05),and the protein expression levels of NLRP3 and GSDMD in the hippocampus significantly increased(P＜0.01).Compared with model group and sham acupuncture group,the body weight,the sucrose preference rate,the horizontal exercise score and the vertical exercise score increased in electroacupuncture group(P＜0.05),and the protein expression levels of NLRP3 and GSDMD decreased(P＜0.05).Conclusion Yishen Jieyu acupuncture combined with electroacupuncture may play an antidepressant role by inhibiting the expression of the pyroptosis-related proteins NLRP3 and GSDMD.关键词
