摘要

Abstract

Objective To investigate the inhibitory effect of the combination of sorafenib and evodiamine on the proliferation of HepG2 cells and the underlying mechanism.Methods The inhibitory effects of sorafenib and evodiamine used alone or in combination on the growth of HepG2 cells were evaluated by CCK-8 assay.The apoptosis levels were detected by flow cytometry using the Annexin V-FITC/PI double staining method.The ex-pression changes of apoptosis-related proteins were detected by Western blot.Results Both sorafenib and evodi-amine had obvious inhibitory effects on the proliferation of HepG2 cells(P<0.05),and the apoptosis rate of the combined treatment group was significantly higher than that of the single treatment group(P<0.05).Com-pared with the single use group,the expression levels of Bax and Cleaved caspase-3 proteins in the combination group were significantly increased,while the protein expression level of Bcl-2 was significantly lower than that in the single use group(P<0.05).Conclusion These results suggested that sorafenib and evodiamine had a syn-ergistic antitumor effect on hepatoma HepG2 cells,and the possible mechanism is related to the induction of tumor cell apoptosis.
