摘要

Abstract

Objective To design and synthesize a fluorescent probe for lysosomal imaging.Methods The compound(E)-3-(2-pyridine-4-vinyl)-1H-indole(PVI)was synthesized by using racholine and 4-pyri-dine formaldehyde as substrates and n-tributylphosphine as catalyst.The structure of the compounds was charac-terized by nuclear magnetic resonance(NMR).The relationship between the probe and the pH of the solution was explored by fluorescence spectroscopy and UV-vis absorption spectroscopy.The mechanism of pH response was estimated by theoretical calculation.The cell imaging was observed by inverted fluorescence microscope.The lysosomal co-localization imaging was recorded by confocal laser scanning microscopy.Results The fluores-cence intensity of PVI was gradually increased with the decrease of pH when the pH range from 4.00 to 7.00.PVI had low cytotoxicity and could be used for imaging PC-3 prostate cancer cells and their lysosomes.Conclu-sion A pH-responsive fluorescent probe PVI has been synthesized,and shows the advantages of low cytotoxicity,good biocompatibility,and high fluorescence intensity.As a lysosome fluorescent probe with,it can be used for lysosome localization and imaging,and has commercial application value.
