Abstract

The method of extracting triterpenoids from alcohol extract of Ganoderma lucidum was studied by constructing the aqueous two-phase system of salt and alcohol.The extraction efficiency of Ganoderma lucidum triterpenes was investigated by an alcohol-salt system consisting of 13 inorganic salts and ethanol.It was found that the EtOH-NaH2 PO4 system had the highest extraction efficiency.The effects of NaH2 PO4 concentration in aqueous solution,mass fractions of ethanol and NaCl,and pH of aqueous phase on extraction efficiency were investigated by single factor experiment.The optimized process conditions were obtained by orthogonal experiment:the mass fractions of NaH2 PO4,water,ethanol and Ganoderma lucidum alcohol extract were 10%,53%,27%and 10%,respectively.Before extraction,solid NaOH was used to adjust the pH of aqueous phase to 5.At the same time,the results of the two methods of extraction,namely,the aqueous two-phase and the alkaline-acid solvent,were also compared.It was found that the extraction rate of triterpenes of Ganoderma lucidum by the aqueous two-phase method was 47.366%,which was higher than that by the alkaline extraction and acid solution(44.933%).Furthermore,the aqueous two-phase extraction was proved to be an efficient and green extraction method.
