黄河兰州段一株优势藻的分离鉴定及光照、pH、温度对其生长的影响
Isolation and identification of a dominant algae in Lanzhou section of the Yellow River and the effects of light,p H and temperature on its growth

Abstract
To explore the dominant green algae in the Lanzhou section of the Yellow River,water samples were collected from the Jinniujie Wharf and Zhongshan Bridge waters,and four microalgae strains were isolated.They were preliminarily identified as Chlorella,Desmodesmus,Raphidocelis and Coelastrum,and were named Chlorella sp.HH13,Desmodesmus sp.HH2C,Raphidocelis sp.HHTXZ and Coelastrum sp.HH4.After 9 d of cultivation,the biomass level of four algae strains showed HH13>HH2 C>HHTXZ>HH4.HH13 was selected as the target algae species to investigate its biomass,photosynthetic pigments,soluble proteins and soluble sugar accumulation under the optimal conditions of light,p H and temperature.Cultivate HH13 in BG11 medium was cultivated and the optimal light intensity of 6000 Lux was determined.The content of carotenoids and total chlorophyll reaches the maximum value,which is 2.029 and 6.929 mg·g-1,respectively.The content of soluble sugars and proteins is also the highest,which is 0.285%and 7.661 mg·g-1,respectively.The optimal p H value is 7,and the maximum biomass can reach 0.297 g·L-1.The content of carotenoids and total chlorophyll reaches the maximum value,which is 2.753 and 9.711 mg·g-1,respectively.The content of soluble sugar and soluble protein is also the highest,which is 0.484%and 5.674 mg·g-1,respectively.The optimal temperature is 28℃,the highest biomass is 0.289 g·L-1,the content of carotenoids and total chlorophyll is 1.796 and 6.073 mg·g-1,respectively.The content of soluble sugar is 0.288%,and the content of soluble protein is 5.898 mg·g-1.The optimal light intensity for the growth of HH13 is 6000 Lux with a p H value of 7 and a temperature of 28℃.关键词
