Abstract

Objective To predict the quality marker(Q-Marker)of Jingu Tongxiao Tincture based on fingerprint and network pharmacology.Methods High-performance liquid chromatography(HPLC)method was used to establish the fingerprints of 17 batches of Jingu Tongxiao Tincture,and the Similarity Evaluation System for Chromatographic Fingerprint of Traditional Chinese Medicine(Version 2012A)was used for similarity evaluation and identification of common components.The network pharmacology was used to screen related targets and key pathways of common components,to establish a"component-target-pathway"network,and to predict potential Q-Markers that play a role in promoting blood circulation,resolving blood stasis,unblocking collaterals,dispelling cold,dispersing nodules,and relieving pain.Results A total of 29 common peaks were calibrated and six chromatographic peaks were identified,namely loureirin B,acetyl-11-keto-β-boswellic acid,asperosaponin Ⅵ,hydroxysafflor yellow A,rhein,and amygdalin.A total of 50 key targets and 90 enrichment pathways were screened out,and based on the connectivity among compounds,targets,and signaling pathways,five Q-Markers of acetyl-11-keto-β-boswellic acid,asperosaponin Ⅵ,hydroxysafflor yellow A,rhein,and amygdalin were preliminarily predicted.Five key targets of albumin(ALB),JUN,tyrosine kinase(SRC protein),prostaglandin-endoperoxide synthase 2(PTGS2),and mitogen-activated protein kinase(MAPK)were calculated based on the protein-protein interaction(PPI)network.Gene Ontology(GO)enrichment analysis involved 197 biological processes(BP),35 cell components(CC),and 64 molecular functions(MF).The Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis obtained highly correlated signaling pathways,including lipid and atherosclerosis,relaxin,interleukin-17(IL-17),osteoclast differentiation,apoptosis,C-type lectin receptor,Th17 cell differentiation and autophagy.Conclusion The established method is easy to operate and has good repeatability.It preliminarily predicts five Q-Markers of Jingu Tongxiao Tincture,which can provide a reference for its quality standards and mechanism of action.
