Abstract

Objective To evaluate the genetic stability of tissue-cultured Viticis Negundo Folium.Methods The extraction effects of four solvents on the total flavonoids from Viticis Negundo Folium were investigated.High-performance liquid chromatography(HPLC)method was used for the content determination of vitexin,and the inhibitory effect of the extract of Viticis Negundo Folium on α-glucosidase and the kinetic curve of vitexin were tested.Results The extraction rates of active ingredients from Viticis Negundo Folium by different solvents were as follows:acidic ethanol(pH 1.33)>70%ethanol>acetone>ethyl acetate.In the tissue-cultured and wild Viticis Negundo Folium,the total flavonoid contents extracted with acidic ethanol as solvent were 62.79 mg/g and 60.35 mg/g(calculated by fresh weight),respectively;the vitexin contents were 132.20 μg/g and 126.12 μg/g(calculated by fresh weight),respectively;the 50%inhibitory concentrations(IC50)of the extract on α-glucosidase were 1.41 mg/mL and 1.37 mg/mL,respectively.The kinetic curve analysis showed that the inhibitory type of the extracts from tissue-cultured and wild Viticis Negundo Folium on α-glucosidase was reversible inhibition.Conclusion The extract of Viticis Negundo Folium has a strong inhibitory effect on α-glucosidase,and the active ingredients of tissue-cultured and wild Viticis Negundo Folium are similar,indicating that tissue-cultured Viticis Negundo Folium has good genetic stability.
