Abstract
The purpose of this experiment was to explore adisinfectant that could efficiently degrade the nucleic acid of pseudorabies virus and simulate its disinfection effect in actual disinfection scenarios in pig farms.In order to guide the prevention and control of pseudorabies in farms and improve their biosafety capabilities.Used pseudorabies virus positive tissue grinding fluid as a disinfection object and used 7.5,15,30 and 60 ppm hypochlorite disinfectant to disinfect at room temperature and 4℃ for 10,15,30 and 45 min,respectively.The effects of immersion(10,15,30,45 min),spray(10,15,30,45 min),wiping(2,5,20,50 times)and high-pressure washing(1,2,5,10 min)on nucleic acid degradation were compared.In addition,for common pig farm materials such as stainless steel,plastic bottles,plastic bags,woven bags,vaccine bottles,cement floors,tiles,newspapers,clothes,egg shells,instant noodles,vegetables,tires,and wooden boards,applied an equal amount of pseudorabies virus positive tissue grinding fluid to the surface.Soaked in an excess of 60 ppm hypochlorite disinfectant for 20 minutes,and used fluorescence PCR method to sample and tested the disinfected surface.The results showed that within 30 minutes of disinfection,only the 60 ppm hypochlorite group tested negative.All ≥30 ppm groups and 15 ppm 45 min groups in ambient environment were negative.All ≥30 ppm groups were negative at 4℃ and nor-mothermic environments.In the disinfection method,all immersion groups,high-pressure groups,spray 45 min groups,wiping 20 times groups,wiping 50 times groups were negative.After 20 minutes of disinfection with excessive 60 ppm hypochlorite on the surface of conventional materials in the breeding farm,the results were all negative.In summary,hypochlorite disinfectant was an efficient disin-fectant that could quickly and efficiently degrade the pseudorabies virus nucleic acid on the surface of common materials in breeding farms when used for flushing and soaking.Low temperature also had a certain inhibitory effect on hypochlorite disinfectant.关键词
