基于PI3K/Akt/NF-κB信号通路探讨加味升降散对糖尿病肾病小鼠肾损伤的干预机制

Abstract
Objective:To investigate the intervention mechanism of Jiawei Shengjiang powder(JSP)on renal injury in diabetic nephropathy mice based on PI3K/Akt/NF-κB signaling pathway.Methods:According to the body mass stratification method,30 db/db mice were randomly divided into a model group,a JSP low-dose group,a JSP medium-dose group,a JSP high-dose group,and a perindopril group,with 6 mice in each group;and 6 db/m mice were taken as blank group.The mice were fed with free diet and water for 12 weeks,and then samples were collected.Serum creatinine(SCr)and blood urea nitrogen(BUN)were detected through fully automatic biochemical analyzer.The 24-hour urinary albumin,NGAL,TNF-α,IL-1β,VCAM-1,MCP-1 and HbA1c were measured by ELISA.Western blot was used to detect the contents of p-PI3K,p-NF-κB p65 and p-Akt.Furthermore,the pathological changes of the kidneys were observed.Results:Compared with model group,in the groups of JSP,SCr and BUN,24-hour urinary albumin,the levels of NGAL and HbA1c,and the expressions of inflammatory factors such as TNF-α,IL-1β,VCAM-1 and MCP-1 of db/db mice were reduced.The expression levels of p-PI3K and p-AKT were up-regulated(P<0.05),and the activation of p-NF-κB p65 protein were inhibited in the groups of JSP(P<0.05).Moreover,pathologically,the damage of renal tubular epithelial cells,glomerular capillaries and basement membrane were improved in the JSP groups,and the damaged podocytes in diabetic nephropathy mice were repaired.Conclusion:Jiawei Shengjiang Powder can reduce kidney inflammation and repair podocyte injury by regulating PI3K/AKT/NF-κB signaling pathway,and therefore,it plays a role in the treatment of diabetic nephropathy.关键词
