桦褐孔菌黄酮对糖尿病肾病小鼠的治疗作用研究
Therapeutic Effect of Inonotus Obliquus Flavonoids on Diabetic Nephropathy in Mice

Abstract
Objective:To study the therapeutic effect of inonotus obliquus flavonoids on diabetic nephropathy(DN)in mice.Methods:50 C57BL/6 mice(4-week-old)were taken,of which 6 were used as the normal control group.The rest mice were fed with streptozotocin(STZ)combined with high glucose and high fat diet for 9 weeks to establish diabetic nephropathy model.After successful modeling,30 mice were randomly divided into a diabetic nephropathy group,a positive control group(valsartan 20 mg/kg),an inonotus obliquus flavonoid low dose group(50 mg/kg),an inonotus obliquus flavonoid medium dose group(100 mg/kg)and an inonotus obliquus flavonoid high dose group(200 mg/kg),with 6 rats in each group.The rats were gavaged for 6 weeks,and after 6 weeks,the changes of immune function were observed.Results:Compared with the normal group,the expressions of MHC-Ⅱ,CD86,CD54 and CD80 on the surface of exosomes secreted by bone marrow derived dendritic cells and their supernatant in mice with diabetes nephropathy were significantly reduced(P<0.01).T lymphocytes of CD3+,CD4+,CD8+ and IFN-γ secreted by spleen decreased(P<0.01).The number of CD3+,CD4+,CD8+ T lymphocytes in the spleen and the IFN-γ factors secreted by these cells were reduced(P<0.01).After the treatment,the expression levels of cell and exosome surface molecules MHC-Ⅱ,CD86,CD54,and CD80 increased in all groups(P<0.05),and the numbers of CD3+,CD4+,CD8+ T lymphocytes and IFN-γ(P<0.05)also increased.Conclusion:Inonotus obliquus flavonoids can improve the immune function of mice with diabetic nephropathy,and have good therapeutic effect on the disease with high safety.关键词
