大黄素骨水泥对感染性创面炎症因子的作用及机制研究
Study on the effect and mechanism of Emodin-loaded bone cement on inflammatory factors in infected wounds

Abstract
Objective To observe the effect of Emodin-loaded bone cement on the repair of infectious wound and to study the mechanism.Methods Sixty rats were divided into normal group,model group,Emodin-loaded bone cement group(PMMA+E)and vancomycin bone cement group(PMMA+V),except for the blank group,a model of total skin excision on the back of rats infected with Staphylococcus aureus was constructed.The number of colonies in each group was measured on days 1,7 and 14 after the intervention,and the histopathological changes of the wounds were observed by HE staining,IL-6 and TNF-α in the serum of rats were detected by ELISA,and IL-6 and TNF-α in the wounds were detected by RT-qPCR.Western-Blot detection of TLR4/NF-κB signaling pathway protein expression levels in wounds.Results On days 7 and 14 after the intervention,the wound area of rats in the PMMA+E group was significantly reduced compared with the model group and PMMA+V group(P<0.05).Compared with the model group,the number of colonies in the PMMA+E group was significantly reduced(P<0.05);compared with the PMMA+V group,there was no significant difference.Compared with the model group and PMMA+V group,HE staining in the PMMA+E group showed a decrease in inflammatory cell infiltration and a large number of new blood vessels.Compared with the model group and PMMA+V group,the serum IL-6 level was significantly lower and TNF-α level was significantly lower(P<0.05)in the PMMA+E group.Compared with the model group,the wound IL-6 mRNA content of rats in the PMMA+E group was significantly lower(P<0.05);compared with the PMMA+V group,there was no significant difference.Compared with the model group and PMMA+V group,the wound TNF-α mRNA content of rats in the PMMA+E group was significantly lower(P<0.05).Compared with the model group,the expression of TLR4 and NF-κB protein in the traumatic tissue of rats in the PMMA+E group was reduced(P<0.05);compared with the PMMA+V group,there was no significant difference.Conclusion Topical emodin can effectively inhibit the growth of Staphylococcus aureus and suppress the serum inflammatory factor level as well as wound inflammatory factor level in rats with infected wounds,thus promoting wound healing,and its mechanism of action may be related to TLR4/NF-κB signaling pathway.关键词
