摘要

Abstract

A strain with high production of endoglucanase was selected from the straw fermentation of cattle stomach in Changsha County,Hunan Province,and was named SCUEC7.Based on the morphological observation,physiological and biochemical characteristics,and 16S rDNA sequence comparison analysis,SCUEC7 strain was identified as Bacillus subtilis.The culture conditions for the better growth of SCUEC7 strain were as follows:12 h of culture period,pH 6.0,37℃of culture temperature,20.0 g·L-1 maltose as carbon source,15.0 g·L-1 tryptone as nitrogen source,1.5 g·L-1 Mn2+ as metal ion.Response surface methodology analysis showed that the highest predicted enzyme activity was 409.0 U·mL-1 when the initial pH value was 5.9,maltose content was 19.8 g·L-1,and culture period was 11.8 h.There was a high degree of fit between the measured and predicted values of endoglucanase activity.SCUEC7 strain has a strong ability to produce endoglucanase,which can provide a basis for the development of animal feed.
