山茶油对H2O2诱导大鼠H9C2心肌细胞氧化应激损伤的保护作用OACSTPCD
Protective effect of camellia oil on H2O2-induced oxidative stress injury in H9C2 cardiomyocytes of rats
目的:探讨山茶油对H2O2 诱导大鼠H9C2心肌细胞氧化应激损伤的保护作用.方法:CCK8法检测不同浓度H2O2 处理H9C2心肌细胞后细胞存活率,以正常培养的细胞为空白对照组,以 200 μmol/L H2O2处理细胞24 h,构建氧化应激损伤模型作为模型组.分别用1%,0.1%,0.01%浓度的山茶油预处理细胞24 h后,加入H2O2作用24 h作为实验组.利用β-半乳糖苷酶衰老染色实验,线粒体膜电位实验,EdU细胞增殖染色实验及划痕实验来观察各组细胞衰老,线粒体膜电位,增殖凋亡,迁移的变化.利用试剂盒检测各组细胞超氧化物歧化酶(SOD)活性、乳酸脱氢酶(LDH)活性、丙二醛(MDA)含量.结果:不同浓度H2O2处理H9C2心肌细胞后,细胞活力均受到抑制,且与H2O2浓度正相关(P<0.01).与空白对照组相比,H2O2模型组细胞衰老阳性率、MDA含量和LDH活性增加(P<0.01);线粒体膜电位、细胞增值率、迁移率、SOD活性下降(P<0.01).与H2O2 模型组相比,实验组细胞衰老阳性率(P<0.05)、MDA含量和LDH活性降低(P<0.05);线粒体膜电位增加、细胞增殖率和迁移率增加(P<0.05).结论:山茶油可显著抑制H9C2细胞的氧化应激损伤,发挥心肌细胞保护作用.
Objective:To investigate the protective effect of camellia oil on H2O2-induced oxidative stress injury in H9C2 cardiomyocytes.Methods:The survival rate of H9C2 cardiomyocytes treated with different concentrations of H2O2 was detected by CCK8 method.Normal cultured cells were treated with 200 μmol/L H2O2 for 24 h,and the oxidative stress injury model was established as the model group.Cells were pretreated with 1%,0.1%,0.01%concentration of camellia oil for 24 h,and H2O2 was added for 24 h as the experimental group.The changes of cell senescence,mitochondrial membrane potential,proliferation,apoptosis and migration in each group were observed by β-galactosidase senescence staining,mitochondrial membrane potential staining,EdU cell proliferation staining and scratch staining respectively.The activity of superoxide dismutase(SOD),lactate de-hydrogenase(LDH)and malondialdehyde(MDA)in each group were detected by the kit.Results:The viability H9C2 cardiomyo-cytes treated with different concentrations of H2O2 inhibited and positively correlated with H2O2 concentration(P<0.01).Com-pared with the blank control group,the positive rate of cell senescence,MDA content and LDH activity in the H2O2 model group were increased(P<0.01).Mitochondrial membrane potential,cell proliferation rate,mobility and SOD activity were decreased(P<0.01).Compared with the H2O2 model group,the senescence positive rate(P<0.01 or P<0.05),MDA content and LDH ac-tivity were decreased in the experimental group(P<0.01 or P<0.05).Mitochondrial membrane potential increased,cell prolifera-tion rate and mobility were increased(P<0.01 or P<0.05).Conclusion:Camellia oil can significantly inhibit the oxidative stress injury of H9c2 cells and play a protective role in cardiomyocytes.
