Effect research of protoopioid on apoptosis,inflammation and oxidative stress of intestinal epithelial cells induced by lipopolysaccharide

Abstract
Objective To investigate the effects of protoopioid on inflammation,apoptosis and oxidative stress of intestinal epithelial cells induced by lipopolysaccharides(LPS).Methods NCM460 cells were screened by methyl thiazolyl tetrazoli-um(MTT)assay and treated with 5 μg/ml LPS,and the cells were stimulated with different concentrations of protoopioid(0,5,10,20 μM)for 24 h.The viability and apoptosis rate of NCM460 cells were detected by MTT assay and flow cytometry.The expression levels of apoptosis-associated protein B-celllymphoma-2(Bcl-2)and Bcl-2-associated X protein(Bax)in each treatment group were detected by Western blot.The expression levels of inflammatory factors tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β)and interleukin-6(IL-6)were detected by ELISA.The expression levels of superox-idedismutase(SOD),malondialdehyde(MDA),total antioxidant capacity(T-AOC)and myeloperoxidase(MPO)were detected by ELISA and the expression levels of reactive oxygen species(ROS)were detected by flow cytometry to determine the effect of protoopioid on the oxidative stress response of NCM460 cells.Results The viability of NCM460 cells in the 5 μg/ml LPS group was lower than that in the negative control group,the difference was statistically significant(P<0.05).The cell viabil-ity and Bcl-2 protein of NCM460 in LPS+ protoopioid group were higher than those in LPS group in a dose-dependent manner,with statistical significances(P<0.05).The expression levels of apoptosis rate,Bax protein,TNF-α,IL-1β and IL-6,oxidative stress molecules SOD,MDA,T-AOC,MPO and ROS in NCM460 cells in LPS+ protoopioid group were low-er than those in LPS group in a dose-dependent manner,with statistical significances(P<0.05).Conclusion Protoopioid can ameliorate LPS-induced inflammatory injury of intestinal epithelial cells by decreasing apoptosis,inflammatory response and oxidative stress response.关键词
