Effects of ABA Treatment on Postharvest Ripening and Key Genes of Actinidia chinensis cv.'Hongyang'

Abstract
[Objective]The aim of this study was to investigate the effects of abscisic acid(ABA)treatment on postharvest fruit ripening and fruit quality of kiwifruit,thus providing theoretical basis and reference for precise regulation of postharvest ripening process and prolonging storage period of kiwifruit.[Method]In present study,'Hongyang'was used as the experimental material.The kiwifruit clusters were treated with 100 µmol/L ABA and 200 µmol/L nordihydroguaiaretic acid(NDGA,an inhibitor of ABA synthesis)for fruit soaking after harvesting,and water treatment was used as the control.Postharvest fruit respiration intensity,weight loss,hardness,soluble solids,titratable acid,vitamin C and soluble sugar contents were measured to investigate the effects of ABA treatment on the storage quality of kiwifruit fruit.Endogenous ABA content and ethylene release were detected,the expression of key genes of ABA metabolism(AcNCEDs and AcCYP707As)and core component genes of ABA signaling(AcPYLs,AcPP2Cs and AcSnRK2s),as well as the transcriptional expression differences of ethylene-related and cell wall degradation-related genes were analyzed by qRT-PCR.[Result]During the storage period at room temperature,fruit hardness of kiwifruit fruits gradually decreased,and the hardness of all ABA-treated groups were lower than that of the control group,while at the 8th day of storage,ABA significantly increased the expression of cell wall degradation-related genes AcPL1,AcPL2,AcXTH5,AcXTH6,AcPME1 and AcMAN1,which were 3.01,2.42,7.58,21.47,5.11 and 9.49-fold,respectively,indicating that ABA treatment may promote fruit softening by inducing the expression of genes related to cell wall degradation.Exogenous ABA promoted endogenous ABA biosynthesis and ethylene release in kiwifruit.Tthe expression patterns of AcNCED1 and AcNCED2 were consistent with the trend of ABA content,while the expression analysis of ethylene-related genes revealed that ABA treatment promoted the up-regulation of ethylene synthesis and signal transduction genes.In addition,ABA treatment promoted the degradation of titratable acid and vitamin C,the accumulation of soluble solids,total sugars and soluble sugars,and increased the fruit respiration rate and weight loss rate.These results suggest that ABA promoted postharvest ripening of kiwifruit and affected fruit quality.[Conclusion]Exogenous ABA treatment promotes postharvest ripening of kiwifruit fruit,stimulats the expression of ethylene synthesis and signaling-related genes,as well as cell wall degradation-related genes,and accelerates fruit softening.The results provide a basis for revealing the molecular mechanism of ABA-promoted postharvest ripening in kiwifruit.关键词
