摘要

Abstract

To preserve the resources of Oryza granulata,two kinds of O.granulata introduced from the International Rice Research Institute were selected to induce callus and establish tissue culture rapid propagation system,and the callus tissue was preserved at ultra-low temperature.The results showed that the calli of wild rice could obtained by seed induction and radicle induction after germination.The alternative subculture of NB and MS medium was helpful to improve the callus activity.By the combination of high sugar pre-culture and drying treatment,the calli of two kinds of O.granulata survived after cryopreservation,with callus proliferation rates of 34.24%and 23.95%,and the regenerated calli could differentiate into new plants.The combination of tissue culture and rapid propagation technology with cryopreservation provided an effective way for the preservation of O.granulata germplasm.
