Establishment of a bone marrow-derived cell-specific gene-modified mouse model by bone marrow transplantation

Abstract
Objective:To establish a mouse model of bone marrow-derived cell-specific gene modification by bone marrow transplantation.Methods:Bone marrow cells from donor mice(C57BL/6 wild-type)with the same genetic background were transplanted into X-ray irradiated recipient mice(C57BL/6 Ep3-/-mice),and the optimal irradiation regimen for the bone marrow transplantation experiments in genetically modified mice was determined by the comparison of different radiation dosages and modalities.Blood and tail tissue DNA of recipient mice were extracted for genotype identification and chimerism rate analysis.Results:After recipient mice were irradiated by X-rays with appropriate dose(radiation dose of 8.0 Gy,irradiated in 2 times,4.0 Gy each time,with an interval of 6 h),the survival rate of the mice could reach(74.3±2.4)%and the chimerism rate(95.77±0.52)%,which was in accordance with the requirements of bone marrow transplantation experiments.Conclusion:A bone marrow-derived cell-specific gene-modified mouse model that meets the requirements of bone marrow transplantation experiments has been successfully established,which can be used for cardiovascular and inflammation-related research work.关键词
