基于网络药理学和分子对接探讨鸡血藤治疗糖尿病肾病的作用机制研究
Study on the mechanism of Caulis spatholobi in the treatment of diabetes kidney disease based on network pharmacology and molecular docking

Abstract
Objective To explore the potential mechanism of Caulis spatholobi in the treatment of diabetic kidney disease(DKD)based on network pharmacology and molecular docking.Methods The active components and core targets of Caulis spatholobi were selected by searching several databases,and DKD targets were collected by Drugbank,TTD,OMIM,Gene-card and PharmGkb databases,and Venny 2.1.0 was used to obtain the intersection target genes of Caulis spatholobi and DKD.Protein-protein interaction(PPI)network was constructed by String database,and Cytoscape software and CytoNCA plugin were used to analyze and screen out core targets.And then,gene ontology(GO)functional enrichment analysis and Kyoto encyclopedia of genes and genomes(KEGG)pathway analysis were performed.Finally,AutoDock Tools software was used to verify the binding ability of the active compounds to the core targets by molecular docking technology.Results 24 key active components and 12 818 DKD targets of Caulis spatholobi were excavated,with 48 intersecting target genes of the two.3 active components of Caulis spatholobi(aloe-emodin,luteolin,licochalcone A)and 4 core target genes(heat shock protein 90 alpha family class A member 1[HSP90AA1],MYC proto-oncogene[MYC],tumor protein p53[TP53],signal sensors and transcriptional activator 3[STAT3])were screened.A total of 498 entries were enriched by GO function,and 61 entries were enriched by KEGG pathway.Molecular docking technology had verified that three active compounds had reli-able affinity for their key targets of action.Conclusion It is inferred that Caulis spatholobi has the characteristics of multi-component,multi-target and multi-pathway in the treatment of DKD,which provides a new idea for the further study of the molecular mechanism of Caulis spatholobi in the treatment of DKD.关键词
