Abstract
AIM To establish a simple,rapid and efficient method for the determining of voriconazole concentration in human plasma,and apply it to the individualized use of voriconazole.METHODS After protein precipitation with acetonitrile,using carbamazepine as internal standard,plasma concentration of voriconazole were determined by HPLC method.The determination was performed on Waters T3 column(250 mm × 4.6 mm,5 μm)with mobile phase consisted of methanol-water(63∶37)at the flow rate of 1.0 mL·min-1.The detection wavelength was 256 nm,and the column temperature was maintained at 30℃.The sample size was 20 μL.Meanwhile,the method was applied to determine plasma concentration of voriconazole in 20 patients receiving voriconazole.For patients with substandard blood drug concentration of voriconazole,analysis was conducted based on genotype and concomitant medication,and intervention suggestions were proposed.RESULTS A good liner relationship of voriconazole was obtained within the range of 0.5-15 mg·L1.The lower limit of quantitation was 0.5 mg·L-1.The extraction recoveries were 85.71%-93.85%.RSD of intra-day was lower than 7%and RSD of inter-day was lower than 8%.The stability tests was lower than 10.2%.The plasma concentration of voriconazole was 0.57-7.87 mg·L-1 in 20 patients.Among them,6 patients were not within the effective concentration range.After adjusting the medication regimen for 48 h,the concentration of voriconazole were retested to be within the effective concentration range.CONCLUSION The study establishes a simple,rapid and efficient HPLC method for the determination of voriconazole in human plasma,and the clinical therapeutic effect can be improved by monitoring the plasma concentration.关键词
