Effects of Panobinostat on proliferation,apoptosis,and metastasis of human anaplastic thyroid carcinoma ARO cells
摘要
Abstract
Objective To explore the effects and possible mechanisms of Panobinostat on the proliferation,apoptosis and metastasis of human anaplastic thyroid carcinoma ARO cells.Methods ARO cells were cultured in vitro and divided into a control group and Panobinostat low,medium,and high dose(2,4,8 μmol/L)groups for testing.MTT assay was used to detect the inhibitory effect of Panobinostat on the proliferation of ARO cells.Scratch assay was used to detect the effect of different concentrations of Panobinostat on ARO cell migration.Flow cytometry was used to detect cell apoptosis.real-time quantitative PCR(RT-qPCR)and Western blot were used to detect the influence of gene and protein levels of B-cell lymphoma-2(BCL-2)-associated X protein(BAX),BCL-2 and matrix metalloproteinase-9(MMP9).Results The MTT results showed that compared with the control group,Panobinostat had a significant inhibitory effect on the proliferation of ARO cells in a concentration-dependent manner(P<0.05).The results of scratch assay showed that compared with the control group,the migration ability of ARO cells decreased with the increase of drug concentration,with a statistically significant difference(P<0.05).The results of flow cytometry showed that compared with the control group,the apoptosis rate of ARO cells increased with the increase of drug concentration,with a statistically significant difference(P<0.05).RT-qPCR and Western blot results showed that compared with the control group,the gene and protein expression levels of BAX in 2,4 and 8 μmol/L groups were increased(P<0.05),while the gene and protein levels of BCL-2 and MMP9 were decreased(P<0.05).Compared with 2 and 4 μmol/L groups,the gene and protein expression levels of BAX in the 8 μmol/L group were increased(P<0.05),and the gene and protein levels of BCL-2 and MMP9 in this group were decreased(P<0.05).Conclusion Panobinostat can promote apoptosis of ARO cells,thereby inhibiting cell proliferation and metastasis.关键词
