电针对脑缺血再灌注大鼠内质网应激IRE1 α/TRAF2/ASK1/Caspase-12通路的影响研究
Effect of EA on IRE1α/TRAF2/ASK1/Caspase-12 Pathway under Endoplasmic Reticulum Stress in CIRI Rats

Abstract
Objective:To observe the effect of electro-needling Baihui(DU20)and Neiguan(PC6)on apoptosis related proteins of IRE1α,TRAF2,ASK1 and Caspase-12 in ischemic brain tissues of rats with cerebral ischemia-reperfusion injury(CIRI),thus to explore the specific mechanism of electro-acupuncture(EA)in treatment of CIRI.Methods:60 male SD rats were randomly divided into the sham operation group,the model group,the acupuncture group and the Edaravone group,with 15 rats in each group.The model of middle cerebral artery occlusion(MCAO)was established by using modified suture method.LONGA five-grade neurological function score was used to evaluate neurological function.HE staining was used to observe pathological changes of ischemic brain tissues.TUNEL method was used to detect apoptosis rate of neurons.The protein expressions of IRE1α,TRAF2,ASK1 and Caspase-12 in ischemic brain tissues were detected by Western blotting method.Results:Compared with those in the sham operation group,the the neurological deficit score,the apoptosis rate and the protein expressions of IRE1α,TRAF2,ASK1 and Caspase-12 were significantly increased(P<0.01);the cortex tissues were disorderly arranged with atrophic and necrotic cells in the model group.Compared with those in the model group,the neurological deficit score,the protein expressions of IRE1α,TRAF2,ASK1 and Caspase-12 were significantly decreased(P<0.01),and the apoptosis rate was also significantly decreased(P<0.01,P<0.05);the pathological condition of cortex was alleviated with less atrophic and necrotic cells in the acupuncture group and the Edaravone group.There were no statistical differences in the above indexes between the acupuncture group and the Edaravone group(P>0.05).Conclusion:EA can inhibit the apoptosis of brain tissues in CIRI rats,and promote the recovery of neural function,which may be related to down-regulating the proteins of IRE1α/TRAF2/ASK1/Caspase-12 pathway.关键词
