Abstract
Objective To explore the effect of dihydroartemisinin(DHA)on apoptosis of human myelodysplastic syndrome(MDS)cells and its mechanism.Methods Human MDS SKM-1 cells were treated in vitro with DHA,or in combination of apoptosis inhibitors or with chemotherapeutic drugs.The cell viability was detected by cell counting kit-8(CCK-8)method;the cell survival status was detected by propidium iodide(PI)staining;the intracellular reactive oxygen species(ROS)level was determined by flow cytometry.Results CCK-8 assay showed that DHA inhibited the proliferation of SKM-1 cells in a concentration-dependent manner;and the effect of DHA was blocked by apoptosis inhibitors,which was also confirmed by PI staining.After SKM-1 cells were treated by DHA plus chemotherapy drugs 5-AZ or Ara-C,the cell survival rate was significantly lower than that treated by single drugs.The flow cytometry showed that with the increase of drug concentration,the apoptosis rate and ROS levels increased.After SKM-1 cells were treated by DHA plus5-AZ or Ara-C,the ROS levels and apoptosis rate were more markedly.Conclusion DHA can induce apoptosis of SKM-1 cells in a concentration-dependent manner,generate ROS,and have a synergistic effect with 5-AZ and Ara-C.关键词
