Construction of a prognosis-predicting model for acute myeloid leukemia based on copper metabolism-related genes

Abstract
Objective To construct a prognosis-predicting model for acute myeloid leukemia(AML)based on copper metabolism-related genes.Methods The gene expression data and corresponding clinicopathological characteristics of AML patients were downloaded from The Cancer Genome Atlas(TCGA)and Therapeutically Applicable Research to Generate Effective Treatments(TARGET)databases.Univariate Cox analysis and LASSO regression were used to screen copper metabolism-related genes associated with AML prognosis,and a prognosis-predicting model for AML was constructed.According to the risk scores of the model,patients were divided into high-risk and low-risk groups.Kaplan-Meier survival curve and receiver operating characteristic(ROC)curve were used to analyze the prognostic value of the model.Subsequently,a nomogram based on the risk score of the model was developed and its predictive performance was verified.Immune infiltration analysis was used to study the characteristics of the tumor microenvironment associated with risk scores.Results The genes related to copper metabolism were screened in databases,16 of which were associated with the prognosis of AML.Age,FAB subtype and cytogenetic classification were associated with high-risk scores of the model.The area under the ROC curve(AUC)of the model in predicting 1-,2-and 3-year survival of AML patients was 0.899,0.890 and 0.951,respectively.Kaplan-Meier survival curve showed that the overall survival of patients with high-risk scores was significantly shorter than that with low-risk scores(P<0.01).The high-risk score of the model was associated with infiltrating immune cells(M2 macrophages,regulatory CD4+T cells,and NK cells).Conclusion The prognosis-predicting model constructed based on copper metabolism-related genes can effectively predict the overall survival of AML patients.The risk score is related to the age,FAB subtype and cytogenetic classification of patients.关键词
