为建立检测鸡产蛋下降综合征病毒(EDSV)的间接免疫荧光方法,并对该检测方法的各个步骤进行优化,以鸡产蛋下降综合征病毒多抗血清和FITC标记的兔抗鸡IgG为组合,对一抗和二抗的最适工作浓度和孵育时间进行了优化,对IFA方法进行了敏感性、特异性和重复性试验.结果表明,检测接种EDSV的96孔细胞培养板时,多抗血清的最适工作浓度为1∶320,孵育时间为45 min;FITC标记的兔抗鸡IgG的最适工作浓度为1∶200,孵育时间为30 min;病毒敏感性试验表明,该方法可以检测到10-2TCID50/mL的病毒;多抗血清敏感度试验表明,当多抗血清稀释到1∶1 280时仍可见较明显的特异性荧光;特异性试验表明,该方法只针对EDSV,而不与禽副流感病毒4型、鸡新城疫病毒等其他病原反应;批内重复性试验和批间重复性试验结果表明,该方法具有较好的重复性和可靠性.结果表明,建立的间接免疫荧光方法具有较高的灵敏度和较好的稳定性,可为我国禽源性生物材料EDSV的检测提供技术支持,有巨大的市场需求.

To establish an indirect immunofluorescence assay for the detection of Egg drop syndrome virus(EDSV)in chicken,the various steps of the assay were optimized.In this study,the combination of polyan-tiserum of chicken egg drop syndrome and FITC-labeled rabbit anti-chicken IgG was used to optimize the optimal working concentration and incubation time of the primary and secondary antibodies,and the sensi-tivity,specificity and repeatability of indirect IFA method were tested.The results showed that the optimal working concentration of polyclonal antiserum in 96-well cell culture plates inoculated with EDSV was 1∶320,the incubation time was 45 min.The optimal working concentration of FITC-labeled rabbit anti-chick-en IgG was 1∶200,incubation time was 30 min.Virus sensitivity test showed that the method could detect 10-7.0 TCID50/0.1 mL.The polyclonal antiserum sensitivity test showed that the specific fluorescence was still obvious when the polyclonal antiserum was diluted to 1∶1 280.Moreover,the generated polyclonal an-tiserum against EDSV only targeted at EDSV,did not show cross-reaction with 8 other avian pathogens(FAdV 4,NDV,IBDV,IBV,REV,ALV,MDV,AIV).The results of intra-batch and inter-batch repeat-ability tests showed that the method has good repeatability and reliability.Overall,the preliminary estab-lishment of indirect immunofluorescence assay in this stusy has high sensitivity and stability,also provides technical support for the detection of poultry biological material EDSV and has an important market de-mand.
