摘要

Abstract

Objective:To investigate the effect of Xiongshao decoction(XSD)against liver fibrosis in rats caused by Concanavalin A(ConA)and its effect on TGF-[31/p38MAPK signaling pathway.Method:Totally 10 rats were randomly selected from 50 Wistar male rats as normal group,and the other 40 rats were induced liver fibrosis model by tail vein injection of ConA-PBS solution.The 38 liver fibrosis model rats were randomly divided into model group(n=10),XSD group(n=10),modified XSD group(n=8),Fuzheng Huayu capsule(扶正化瘀胶囊)group(n=10).The pathological changes of liver tissue was observed according to HE staining and Masson staining,and liver function indexes(AST,ALT),liver fibrosis(HA,LN,PC Ⅲ,Ⅳ-C),and hydroxyproline(HYP)in liver tissue were detected.The levels of serum α-SMA and inflammatory factors(TNF-α,IL-1,IL-10)were detected by ELISA,and the relative expression of TGF-β1 and p38MAPK proteins was detected by Western blotting.The p38MAPK mRNA levels in rat liver tissues were detected by RT-qPCR,and TGF-β1 and NF-KBp65 proteins were detected in liver tissues by immunohistochemistry.Results:Inflammatory cell infiltration,abnormal hepatocytes structure,fibrosis and abnormal deposition were seen in the liver tissue of rats in the model group.The degree of liver fibrosis in rats has improved;Liver inflammation has decreased,and collagen fiber proliferation has decreased in all treatment groups.Compared with the normal group,the serum levels of AST,ALT,four indicators of liver fibrosis,α-SMA,TNF-α and IL-1 significantly increased in model group,and the HYP content,TGF-β1,p38MAPK,NF-KBp65 protein expression and p38MAPK mRNA levels in liver tissue significantly increased(P<0.01).Compared with the model group,the serum AST,ALT,four indicators of liver fibrosis,α-SMA,TNF-α and IL-1 levels significantly decreased in each treatment group,and the HYP content,TGF-β1,p38MAPK,NF-KBp65 protein expression and p38MAPK mRNA levels of liver tissue significantly reduced(P<0.01).Compared with Fuzheng Huayu capsule group,TGF-β1 protein expression significantly decreased in XSD group and modified XSD group(P<0.05).Except for TGF-β1 index,there was no significant difference in above indicators among the treatment groups(P>0.05).There was no difference in serum IL-10 expression among the groups(P>0.05).Conclusion:XSD shows effect of anti-ConA-induced liver fibrosis and improvement of liver function in rats,and the anti-liver fibrosis mechanism may be related to inhibiting HSCs activation,reducing inflammatory response,and inhibiting TGF-β1/p38MAPK signaling pathway.The modified XSD and XSD show better effect than Fuzheng Huayu capsules on inhibiting the expression of TGF-β1 protein in liver tissues,and there was no obvious difference in anti-liver fibrosis effect and mechanism among XSD,modified XSD and Fuzheng Huayu capsules.
