转录调节蛋白TP53基因敲除增加斑马鱼耐寒能力与运动行为的研究
Knockout of Transcriptional Regulatory Protein TP53 Increases Cold Tolerance and Athletic Ability of Zebrafish

Abstract
Transcriptional regulatory protein TP53(Tumor protein 53)regulates a variety of crucial biological processes including cell cycle,DNA damage,repair,apoptosis,and so on.As a regulatory factor,tp53 is induced under various stresses.However,the role of tp53 under cold stress in fish still remain elusive.In our study,mRNA level of tp53 in zebrafish(Danio rerio)during cold treatment at 8℃was examined,and the effect of tp53 knockout on cold tolerance and athletic ability of zebrafish was investigated by using a tp53 knockout model(tp53-/-).RT-qPCR showed that tp53 mRNA level in larvae and gills increased significantly after cold exposure.Cold tolerance test showed that the knockout of tp53 had enhanced cold tolerance,indicated by significantly increased LT50(lethal time to 50%mortality)(P<0.000 1)at 8℃for cold treatment.Behaviors test showed that the moved distance,average speed and mobility were significantly decreased in both WT(Wild Type)and tp53-/-zebrafish at 18℃,compared with those at 28℃.However,the athletic ability of tp53-/-zebrafish was significantly higher than WT zebrafish including moved distance,average speed and mobility at both temperatures.This work indicates the involvement of tp53 under cold stress,and enhancement of cold tolerance and athletic ability by the knockout of tp53 in zebrafish.The findings provide new insights for further understanding the mechanism of cold tolerance and behaviors in fish.关键词
